Text: Calpain is an intracellular, nonlysosomal cysteine protease whose activity is regulated by calcium. Micro- and millicalpains (CAPN1, OMIM:114220, and CAPN2, OMIM:114230, respectively), which differ in their calcium requirements for protease activity, are ubiquitously expressed. Other calpains are expressed in specific tissues, such as CAPN3 (OMIM:114240), which is predominant in skeletal muscle. Many gene products associated with carcinogenesis are substrates of calpain family enzymes, including FOS (OMIM:164810), JUN (OMIM:165160), p53 (OMIM:191170), and ESR1 (OMIM:133430) (Huang and Wang, 2001).
